Pharmacodynamics
Pharmacodynamics is the study of what a drug does to the body — how it binds receptors and produces its therapeutic and adverse effects. It complements pharmacokinetics, which describes what the body does to the drug.
Pharmacodynamics is the branch of pharmacology that describes what a drug does to the body: the biochemical mechanisms by which it produces its effects and the relationship between drug concentration and response. The classic memory device pairs it with its counterpart — pharmacokinetics is what the body does to the drug (absorption, distribution, metabolism, excretion), while pharmacodynamics is what the drug does to the body.
Most drugs act by binding to targets such as receptors, enzymes, ion channels, or transporters. An agonist binds a receptor and activates it, mimicking the natural ligand; an antagonist binds without activating, blocking the natural signal. Two properties describe the interaction: affinity (how tightly the drug binds) and efficacy (how much response it produces once bound). Dose-response curves capture these relationships, defining potency (the dose needed for a given effect) and maximal effect.
Pharmacodynamic concepts also explain drug safety. The therapeutic index compares the toxic dose to the effective dose — drugs with a narrow therapeutic index, like warfarin or lithium, demand close monitoring. Pharmacodynamic drug-drug interactions occur when two drugs affect the same system, producing additive, synergistic, or antagonistic effects — for example, combining two sedating medications, or using naloxone to reverse an opioid.
